ID 125: Brain Cleaning in OSA

About the study

This study aims to investigate whether there is a change in the brain's waste management system in patients who have severe obstructive sleep apnea (OSA).
It will look at how the levels of certain markers of dementia change in the blood over a 5-week study period.
Currently, continuous positive airway pressure (CPAP) therapy is the gold standard clinical treatment for OSA, and the research team aims to monitor changes in markers for dementia after 2 weeks of usual CPAP treatment.
(when the patient has minimal to no symptoms of OSA) and after 2 weeks of having no CPAP treatment (when the patient's OSA will return).

What's involved

Eligible Participants will undergo two overnight sleep sessions and one medical check up visit.
Participants will be required to undergo a HD-EEG (Sleep Study) overnight with a cannula inserted in the arm for (24hrs) for hourly blood collection.
During each overnight treatment session, sleep, functioning and blood collection will be completed.
Each overnight treatment session will be separated by two-weeks. Participants will be reimbursed for their time.

Visits required

Participants are required to attend the Woolcock Institute for three separate visits.

Eligibility criteria

  • Adult males and females aged 35-65 years
  • Polysomnography-confirmed severe OSA with apnea hypopnoea index (AHI) ≥ 30/hr, with Non-Rapid Eye Movement (NREM) AHI ≥ 15/hr, within the past 2 years
  • Established CPAP use for treatment of OSA with compliance of > 3 months, with ≥ 5 hours use per night for > 5 nights per week
  • Willing to withdraw from CPAP use for 14 nights
  • Able to give informed verbal and written consent
  • Fluent in spoken, and comprehension of English
